Tranqwin is a polyherbal formulation used to enhance the memory and increase blood circulation in the brain. It includes six herbal ingredients in which piperine and withaferine-A shows the memory enhancing effect. A simple TLC densitometric method has been developed for the quantification of these marker compounds in the poly-formulation using HPTLC. These compounds were quantified and found to be 0.49% and 0.089% of piperine and withaferine-A, respectively. The methods were validated for specificity, linearity, precision, accuracy, LOD, LOQ, robustness and ruggedness. The linearity was found to be in the range of 50-300 ng/spot for piperine and 200-1200 ng/spot for withaferine-A. Accuracy of the method was checked by recovery study conducted by standard addition method and the average percentage recovery was found to be 99.34% and 99.52% for piperine and withaferine-A, respectively. The proposed method is simple, specific, precise, accurate and reproducible and can be used for the routine quality control analysis of formulation containing traditional drug.
